Should the Chromecast X1 Ultra be permanently switched on at the mains supply, even when not in use like overnight for example?

Chromecast devices are designed to be permanently switched on, I leave my devices always on.

I turn off mine whenever I don't want to use it. It won't damage it at all.

When you turn it back on do you get the connect to the mains and not the tv usb graphics...the green tick, red cross ones each time?

Well I used to get it on Chromecast 2 each time I turned it on for 1 second for few years and worked perfectly and still works. Now I use Google TV Chromecast and this stick does not have any ticks while booting and doing same thing by unplugging it from outlet whenever I am not gonna use it (I plug it in and out daily. Like 4-6 times a day and have it for 8 or 9 months and no problems at all.
