I am intending to replace some radiators in my house but to do this I will have to isolate the cold water supply to the house.Does anyone know of any issues that could affect the Nest Learning Thermostat 3rd Generation?

Turning off the cold water supply will not affect the Nest Thermostat.  However, you might wan to turn the Mode to OFF while your boiler if offline.
