My current setup looks like this:
Red = R with jumper to R/C
Orange = O/B
Yellow = Y1
Green = G
Blue = C
White = W2
Black = E with jumper to W2
What is the proper wiring configuration for nest Gen4?
Ive seen that the black wire goes to E but currently heat or A/C isn't kicking on. I have a two-stage convertible air handler with auxiliary heat functionality. My guess is the black wire is the main power source. Should it go in the W1 slot?

Answer for Nest setup
Red = RC

To better assist you, I'd like to know if after wiring the thermostat as you let me know in the post, is your system running properly?
Since the orientation parameter controls when your system switches between heating and cooling, I'd suggest adjusting it from B to O in this case. For instructions, refer to this article: Troubleshoot heat pump issues.
