I have a large open plan room (about 150m2) that contains structures that would block sound. With that in mind, how many nest audios should I buy to fill the entire room with music?

If all your looking for is music, in that large an area, I'm not sure that Nest Audio's would be the best choice. That said start with 4 put them in a group and add as needed.

In addition, you can never go wrong with multiple devices giving you that theater-like experience and at the same time, having a room that contains structures to block sounds with only a single Nest Audio, will give still give you the same great, listening sensation.
