Anyone shed any light on how to connect these wires, tried a number of combos and nothing seems to enable Nest to turn on heating. Reinstalled old Honeywell and works no issue. The loop from L to 1 is also confusing me, and then into 4, but this is a combo system, no separate hot water.

From what we can see in the picture, it seems you have a combi boiler with hot water on demand. The wiring diagram for that can be found at page 20. The Live wire needs to go into L terminal on the Nest Heat Link and from there, a loop/bridge into terminal 2, and the Switch Live/Normally Open wire needs to go into Nest's terminal number 3. Let me know how it went.
