Is this setup compatible with Nest E? And how would wiring work? Do I really have turn off breaker ?

Your current thermostat uses a proprietary interface that none of the Nest thermostats support.
Sometimes you can change the interface at the furnace/air handler to the standard configuration as you do have all of the needed wires. Just look at the terminal block on the control board in the furnace/air handler
