Nest thermostat wiring

I’m replacing my old Honeywell thermostat with a brand new Nest thermostat on the second floor in my house. Above is the configuration of the wiring connection that I found. However, note that the blue wire is connected to O/B and the white wire is not connected at all. I set up the same wire configuration on my new Nest thermostat but I noted that after follow all the setting only Heat is coming out. 
The second picture is the configuration of my first Nest thermostat on the first floor done 2 years ago. Note the wire configuration is different and I have 2 AC units (large and small) for the different floors.

so, Q’s: should I connect the new Nest thermostat (2n floor) using the same configuration I have in my first floor or I have to keep the original wire configuration of picture #1?

Thanks for reaching out to the Google Nest Community Forum. Let's sort this out.

 

To be sure, could you please tell us the type of HVAC system you have? Is this a Zoning system, a Conventional system, or a Heat Pump? If this is a Zoning or Conventional unit, try following the same wiring setup as your other existing Nest Thermostat and activating it with Heating, cooling, and a fan to see if it works. Also, you can share with us a photo of your system's control panel to see how the wires were originally labeled.
